◤反应原理(Test Principle):

本试产品采用双抗体夹心ELISA法。用抗Human LEP抗体包被于酶标板上,实验时样品(或标准品)中的Human LEP会与包被抗体结合。后依次加入生物素化的抗Human LEP抗体和HRP酶标记的亲和素,抗Human LEP抗体与结合在包被抗体上的Human LEP结合,生物素与亲和素特异性结合而形成免疫复合物,游离的成分被洗去。加入显色底物(TMB),TMB在HRP酶的催化下呈现蓝色,加终止液后变成黄色。用酶标仪在450 nm波长处测OD值,Human LEP浓度与OD450 OD值之间呈正比,通过绘制标准曲线计算出样品中Human LEP的浓度。

Intra-assay precision (intra-assay precision): Low, medium, and high levels of Human LEP were measured on a single plate 20 times at each level.

Inter-assay Precision (interassay precision): Samples containing low, intermediate, and high levels of Human LEP were assayed 20 times per plate on 3 different plates.
