胰岛素样生长因子I（IGF1）属于与胰岛素原结构同源的胰岛素样生长因素家族。成熟的IGF是通过非活性前体蛋白的蛋白水解加工产生的，其中含有N端和C端的前肽区域。由70个氨基酸组成的成熟人IGF-I与小鼠IGF-I具有94%的同一性，并表现出跨物种活性。IGF-1结合IGF-IR、IGF-IIR和胰岛素受体，在细胞周期进展、细胞增殖进展中起着关键作用。IGF-1的表达受生长激素的调节。R3 IGF-1是IGF-1的83个氨基酸的类似物，其包含完整的人IGF-1序列，在第三位（即R3）用Arg（R）取代Glu（E），在N末端有13个氨基酸的延伸肽。R3 IGF-1的产生是为了提高生物活性。R3 IGF-1在体外明显比人类IGF-I更有效。
